Abstract

FORMULATION AND CHARACTERIZATION OF FAST DISSOLVING ORAL WAFERS OF AN ANTIEMETIC DRUG PALONOSETRON USING NATURAL POLYMERS

ABSTRACT

Nausea and vomiting are essential protective defense processes by which humans as well as vomit-competent animals tend to avoid ingestion and/or digestion of potentially toxic substances. Administration of antiemetic drug plays important role in controlling emesis. Oral wafers are currently the preferred commercial dosage form for extended transmucosal delivery. Natural polymers are now preferred for preparation of oral wafer as they are non-toxic, economic, safe and devoid of side effects. The aim of this study is formulation & characterization of fast dissolving oral wafers of palonosetron using natural polymers. Oral wafers are fabricated & evaluated for various parameters according to standard procedures. Results showed that thickness of fast-dissolving oral wafers was in the range of 95±3 to 112±5 µm, with the most common thicknesses falling between 0.5 and 5 millimeters. The moisture content of fast dissolving oral wafers was found to be between 1.05-1.82%. The fast dissolving oral wafers found to have tensile strengths ranging from 0.72 to 0.82 kg/cm2. Assay % was in range of 97.78%-98.85%. The in-vitro release study for optimized formulation released up to 98.25% of the active drug within 10 minutes, which is much faster than conventional oral solid dosage forms. Stability studies for Optimized wafer F4 revealed % assay of 99.10, 98.95 & 98.74 for 1 month, 2 month& 3 month respectively. Additionally, the results indicated that the wafer was stable and retained its physical form, even after extended storage. This suggests that the wafer is a viable alternative to traditional oral dosage forms, and could provide an improved patient experience.
